Requirement for expression of WW domain containing transcription regulator 1 in bovine trophectoderm development

چکیده
WW domain-containing transcription regulator 1 (WWTR1) is one of the primary effectors in Hippo pathway, which plays essential roles cell differentiation into trophectoderm (TE) and inner mass lineages at blastocyst stage. However, little known about WWTR1 preimplantation development. The present study aimed to explore significance expression development using an mRNA knockdown (KD) system bovine embryos. We first quantitated protein levels from fertilization proteins gradually shifted extranuclear localization during 16-cell stage nuclear by morula was also transiently upregulated KD efficiently repressed levels. embryos developed rates equivalent those controls, but TE numbers were significantly decreased. Representative TE-expressed genes, including CDX2 IFNT decreased blastocysts. These results provide demonstration that responsible for normal

WWTR1 (WW domain containing transcription regulator 1)
WWTR1 (also called TAZ in publications. Therefore, TAZ is used in the following description) is a WW domaing-containing transcriptional coactivator, which was first identified as a 14-3-3 binding protein. TAZ is the downstream component in the Hippo pathway, and also has been found to interact with different pathways, such as Wnt, TGFbeta, etc. TAZ is involved in mesenchymal stem cell different...
متن کاملMicroRNA-144 inhibits proliferation by targeting WW domain-containing transcription regulator protein 1 in papillary thyroid cancer
Papillary thyroid carcinoma (PTC), the most common histological subtype of thyroid cancer, accounts for between 80 and 90% of all thyroid cancer cases. Previous studies have suggested that microRNAs (miRNAs/miRs) are involved in the development of PTC. The aim of the present study was to investigate whether miR-144 inhibits cellular proliferation in PTC. The expression of miR-144 was detected i...
متن کاملExpression of WW domain–containing oxidoreductase WWOX in pterygium
PURPOSE Pterygium was traditionally regarded as a degenerative disease, but certain characteristics suggest that pterygium is probably premalignant tissue. The human WWOX gene, encoding the WW domain containing oxidoreductase (WWOX, FOR, or WOX1), is a candidate tumor suppressor gene. In this study, we investigated the WWOX gene and protein expression in pterygium. METHODS Pterygium tissues w...
متن کاملWWOX ( WW domain containing oxidoreductase )
WWOX is highly expressed in secretory epithelial cells of reproductive, endocrine and exocrine organs and is expressed in all or most other tissues at a lower level. The protein is not expressed or is expressed at low level in many tumor types, including breast, pancreatic, gastric, hepatocellular, ovarian, lung and prostate cancers. Loss of WWOX expression can be due to inactivation by promote...
متن کاملErratum: WW Domain Containing Transcription Regulator regulates human conjunctiva epithelial cell proliferation via inhibiting TGFβ signaling pathway
PURPOSE To investigate the role of Tafazzin (TAZ) protein in regulating the proliferation of normal human conjunctiva epithelial cells and epithelial cells from pterygium tissue. METHODS Conjunctiva epithelial cells were cultured in keratinocytes growth medium and treated with transformation growth factor β (TGFβ) to analyze the expression and translocation of TAZ protein by immunostaining an...
